一 、科研方向

香烟烟雾提取物致肺泡上皮细胞损伤的分子机制。
DNA甲基化在高血压发病机制中的作用 。
ResearchInterests：
Chronicobstructivepulmonarydisease(COPD)isaworldwideleadingcauseofmorbidityandmortalityanditsprevalenceisstillrising.Itisthereforeimportanttounderstandthedevelopmentofthisdiseaseinordertodevelopstrategiesofprevention,treatment,andcure.Cigarettesmokeextract(CSE)isacomplexmixtureofchemicalscontaininghighlevelsofoxidantsandisthemajoretiologicfactorinthedevelopmentofCOPD,ofwhichemphysemaisamajorcomponent.Amongthevariouscelltypeswhichcomposethelung,theepithelialcellsofthealveolarstructureappeartobeamajortargetforoxidantinjury.ItisnowwellestablishedthattypeIIcellsarethestemcellsofthealveolarepithelium.Afteroxidantinjury,therapidityofinitiationoftypeIIcellproliferationiscrucialforaproperhealing.Therefore,characterizationofthemechanismsinvolvedintheblockoftypeIIcellreplicationbyoxidantsandinitsreversibilityappearstobecriticalfortheunderstandingandmanagementofmanylungdiseasesassociatedwithoxidativestress.Inpreviousstudies,severalcomponentsoftheinsulin-likegrowthfactor(IGF)systemhavebeenreportedinvolvedinthecontrolofproliferationbyusingarattypeIIepithelialcellline,Particularly,accumulationoftheIGFbindingprotein(IGFBP)-2wasassociatedwithblockoftypeIIcellproliferationcausedbyhyperoxia,serumdeprivationandglucocorticoidandsoon.HoweverthemechanismsregulatinghumanalveolartypeIIepithelialcellreplicationuponcigarettesmokeexposureremainpoorlyunderstood.ToprovideinformationontheinfluenceofCSEontherepaircapacityofthealveolarepithelium,IaminterestedinanalyzingtheeffectofCSEontheproliferativeresponseofalveolartypeIIepithelialcells.Todocumentanyprotectiveroleofretinoicacid(RA)toreversetheCSEeffect,wearetoexaminetheconsequenceofRAtreatmentonCSE-inducedgrowthinhibitionoftypeIIepithelialcells
。
AnotherprojectisfocusedontheroleofDNAmethylationinthepathogenesisofhypertension.Accumulatingevidencesuggeststhatkeygenesbecomeepigeneticallyrepressedduringthecourseofnormaldevelopmentandagingaswellasinthecourseofdiseasessuchascancer.itisnotknownwhetherthismechanismisinvolvedinhypertension.Weareintriguedbyourpreliminaryfindingsthatdocumentanup-regulationinexpressionofDNMT1duringgenetic-andangiotensinII(AngII)-inducedhypertension.wespecificallyhypothesizethatDNMTinhibition-mediateddemethylationandre-expressionofeNOSinno-endothelialcellsofthevasculatureresultsinincreasedproductionofbioactiveNO,increasedvasodilation,andloweringofbloodpressure。
